Calcein-AM/EthD-I Dual Staining Assay kit

Calcein AM, EthD-I method Viability/Cytotoxicity Assay Kit for Live & Dead Cells provides dual fluorescence staining for the detection of living and dead cells. The two probes in the kit eflect cell viability by measuring intracellular esterase activity and plasma membrane integrity. This kit can be used for fluorescence microscopes, flow cytometers, microplate readers and other fluorescence detection systems.

Calcein-AM is only used for cell viability testing and short-term labeling of live cells due to the lack of esterase in dead cells. However, Propidium iodide (PI) staining for too long may cause the detected apoptosis rate to be high, while Ethidium bromide dimer-I (Ethidium Homodimer-I; EthD-I) is a high-affinity fluorescent nucleic acid dye with relatively high Strongly positively charged, the dye cannot cross the cell membrane for live cell staining, but can pass through disordered regions of the dead cell membrane to the nucleus and intercalate into DNA double strands to produce red fluorescence. Fluorescence can be enhanced more than 30-fold. Therefore, EthD-I is a more sensitive nucleic acid staining agent for staining mammals, bacteria, yeast and fungi, Ex/Em (binding DNA) = 528/617 nm.
