Total sugar acid is hydrolyzed to reducing sugar. In the presence of NaOH and glycerol, DNS reagent and reducing sugar are heated together and then reduced to amino compounds, which are orange-red in excess NaOH alkaline solution, at 540nm. The maximum absorption peak is used to determine the total sugar content in the sample.
