Mucin stain kit (mild methylation method)
The mild methylation mucin stain is a type of chemical modification and blocking method, mainly used to identify acidic groups in mucins. The principle is to use acidic alcohol solution to eliminate the basophilicity of COOH. When the pH value of Alcian stain solution is 2.5, the carboxyl groups in the tissue are ionized and carry a negative charge, forming salt bonds with the cations in Alcian blue, staining tissues with carboxyl groups (such as proteoglycans/hyaluronic acid and epithelial acidic mucins). It is used untreated sulfated mucins, sulfated proteoglycans, salivary mucins, and hyaluronic acid are blue, and treated sections are light blue.
