Mitochondrial Complex I (EC 1.6.5. 3), also known as NADH-CoQ reductase or NADH dehydrogenase, is widely present in the mitochondria of animals, plants, microorganisms and cultured cells, and is the largest protein in the inner mitochondrial membrane Complex. The enzyme catalyzes the transfer of a pair of electrons from NADH to CoQ, and at the same time reduces O2 to O2.-, which is the main part of the respiratory electron transport chain that produces O2.-. Measuring the enzyme activity can not only reflect the state of the respiratory electron transport chain (ETC), but also reflect the state of reactive oxygen species (ROS) production.
Complex I can catalyze the dehydrogenation of NADH to generate NAD+. The oxidation rate of NADH is measured at 340nm to calculate the enzyme activity.
