Glutamic Pyruvic Transaminase/Alanine Transaminase Activity Assay kit
Glutamic pyruvic transaminase (GPT, EC 2.6.1.2), also known as alanine transaminase (ALT), is widely found in animals, plants, microbes and cultured cells. It catalyzes the reversal of amino reactions and is an important enzyme in amino acid metabolism. In addition, GPT activity is very high in mammalian liver cells. When liver cells undergo necrosis, GPT is released into the bloodstream, resulting in a significant increase in serum GPT activity. Therefore, GPT is recommended as the most sensitive indicator of liver damage by the World Health Organization.
GPT catalyzes the transamination reaction between alanine and alpha-ketoglutarate to generate pyruvate and glutamic acid. The addition of 2,4-dinitrophenylhydrazine solution not only terminates the reaction but also reacts with the carbonyl group in the keto acid to form pyruvate phenylhydrazone, which appears red-brown under alkaline conditions. The enzyme activity of GPT can be calculated by measuring the absorbance at 505 nm.
