DNA End Smoothing Kit
The DNA End Smoothing Kit utilizes the 5'->3' DNA polymerase activity and the 3'->5' exonuclease activity of T4 DNA Polymerase to convert sticky ends produced by restriction enzyme digestion or PCR reactions into blunt ends. The blunt-ended DNA fragments generated by this kit can be used for conventional blunt-end ligation reactions.
There are two types of sticky ends: one with a 5' overhang and the other with a 3' overhang. This kit can fill in the 5' overhangs and also trim (or "chop") the 3' overhangs to create blunt ends. When the end is a 5' overhang, T4 DNA Polymerase can use its 5'->3' DNA polymerase activity to fill in the end; when the end is a 3' overhang, T4 DNA Polymerase can use its 3'->5' exonuclease activity to trim the end.
