Cytochrome P450 enzymes are a group of isoenzymes mainly present in the liver, which play an important role in the metabolism of foreign substances, especially the metabolism of drugs and poisons. Cytochrome P450 and cytochrome b5 are the two heme proteins of the P450 enzyme system, and the change in their ratio is closely related to the metabolic activity of P450.
After oxidized cytochrome b5 is reduced by sodium dithionite, there is a maximum absorption peak at 424nm. The content of cytochrome b5 can be calculated by measuring the difference in absorbance at 424nm and 490nm.
